o Primary contributor in the validation and optimization of the Bionex Dual-Hive automation system for execution of Vividion's proteomic sample prep assay. Created an alternate workflow for troubleshooting the validation of the system by utilizing protein and peptide quantification assays instead of MS analysis to increase throughput of troubleshooting period by 300%.o Developed, optimized, and validated an automated ELISA protocol for application in weekly compound profiling for a lead target. Enabled scientists to increase throughput by 6 hours per week. o Developed, optimized, and validated a VWorks form and 9 protocols to enable fully automated compound plate production for the Platform & Proteomics group. Scripted with JavaScript to allow for combinations of inputs to be captured and used to drive the generation of a runset to produce plates in any desired plate format and dilution scheme. o Execution of Vividion’s proprietary proteomic sample prep assay on a weekly basis to generate MS data enabling the assessment of compound target engagement proteome-wide for new screens and lead evaluation studies. Involved upstream workflow of converting study requests into associated mass-spec experiment ID(s), compound plates, and tracking of experiment progress through the 3-4 day sample prep protocol.o Led Vividion’s design, implementation, and validation of the Bionex Dual-Hive automation system. Involved concept design of a custom dispenser coupled with a vacuum filtration manifold to reduce plate-handling time by 60% for filtration steps during automated runs. Mitigated project challenges involving global supplier shortages and fluid timelines impacting finance decisions over the course of a year.o Developed, optimized, and validated Bravo protocols for all use cases within the Platform & Proteomics group and Biology department, enabling rapid execution of various compound dilution schemes, ELISA(s), and plate reformatting. Protocols were scripted with JavaScript to provide maximum end-user flexibility and ease of use through variable capture and decision trees.o Primary contributor in the management and execution of Vividion’s compound library stamp into mother plates and assay-ready compound plates for use in the proteomic sample prep assay for screening. Developed, optimized, and validated all Bravo protocols and liquid classes used in this process. o Trained end-users to transition assay development and production assays from manual execution to semi- or fully-automated execution to increase throughput and reliability of assay execution. Involved protocol development and validation on platforms like the Agilent Bravo, Formulatrix Mantis, BioTek EL 406, and Bionex Dual-Hive automation system.
