Postdoc ScientistUc Davis Health Mar 2010 - Jun 2015Sacramento, Ca, UsInvestigated the immunity and stress related cell signaling pathways (p38 MAPK, JNK, etc) in Malaria vector mosquito, revealed that inhibition of mosquito p38 or JNK signaling pathways significantly reduced malaria parasite infection, and dissected the detailed mechanism at gene/protein/metabolite levels both in cell and in vivo. • Analyzed mosquito genome sequence and identified 17 MAPKs genes; cloned P38 MAPK and two JNK genes.• Revealed that p38 MAPK take part in the regulation of mosquito anti-malarial peptides through Lucifarase reportor assay in cells and qPCR assay in vivo. • Performed the proteomics experiment (LC-MS/MS) to study the function of mosquito p38 MAPK pathways during infection. The result indicated that p38 MAPK inhibition decreased the antioxidant defense, the mitochondrial protein synthesis, OXPHOS and the protein translation.• Revealed ROS level increased after p38 inhibition with cofocal fluorescence microscopy, and further study indicated that p38 inhibition reduced mosquito’s survival.• Testify the metabolism regulation by p38 inhibition through metabolite analysis on Pyruvate/Lactate level. • JNK function was studied through recombinant expressing two mosquito JNK genes ( with vector pcDNA3.1) in mammalian CHO cell and insect ASE cell; and also using the RNAi (Morpholino) in vivo.• Cooperated with colleagues studying IGF (Insulin-like growth factor) signaling and PKC (Protein kinase C) signaling in malaria vector mosquito. -
Postdoctoral ScholarUniversity Of New Mexico Sep 2008 - Feb 2010Albuquerque, Nm, Us• Recombinant expressed a fibrinogen-related protein (FREP4) from Schistosome vector snail – including the entire protein and the two domains (Immunoglobulin, FBG) in yeast expression system(pPICZα vector and Pitch pastoris strain X-33), and purified the recombinant proteins through affinity chromatography, analyzed their bioactivities on inhibiting bacteria, binding Schistosome SEP, and killing Schistosome in vitro.• To enhance the expression of the entire FREP4 protein, several strategies were applied: 1) try other yeast strains GS115 and KM71H, 2) synthesized a new gene sequence with best synonymous codon bias to Pitch pastoris yeast to let the gene better expressed, 3) transform the yeast with 4X or 8X target genes in one vector, 4) express the protein through different vectors such as the pET32a, pGEX-4T-1, p-ColdI-GST in bacterial expression strain BL21(DE3)plysS, and finally found a way to express enough soluble recombinant FREP4 protein. -
