    My current research focuses on Shh signaling inactivating the tumor suppressor Retinoblastoma (Rb), inducing the Rb target E2F1 and gene regulator peroxisome proliferator-activated receptor γ (PPARγ) in cerebellar neural progenitor cells and in mouse medulloblastoma models. We find Shh-mediated Rb inactivation and E2F1/PPARγ induction promote cancer cell metabolism via glycolysis and mitochondrial fatty acid synthesis, generating a bioenergetic environment permissive for rapid proliferation in progenitor and tumor cells. We are currently investigating pharmacological targets to inhibit tumor growth through cell metabolism in medulloblastoma.

    I have initiated several research projects aimed at evaluating the mTOR pathway, which is important for cell growth and protein translation, in an effort to identify potential targets for therapeutic intervention. These studies primarily involved on understanding the heterotypic crosstalk operating between Sonic hedgehog signaling and the mTOR pathway in the childhood brain tumor medulloblastoma, and have led to the identification of critical mediators specific for these tumor cells. Working with similar cellular systems, recent studies I conducted also led to dual roles of tumor suppressor p27Kip1 in medulloblastoma based on cellular localization: nuclear tumor suppressor and cytoplasmic oncogene.

    My graduate Ph.D. thesis has led to numerous findings:1) 15-Lipoxygenase 2, an arachidonic acid (AA)-metabolizing enzyme, is involved in controlling cell proliferation in normal human prostate (NHP) progenitor cells and prostate cancer cells via AA-dependent and AA-independent metabolic pathways. 2) Immortalized human prostate epithelial cells via hTERT overexpression and inhibition of p16/Rb pathway leads to normal development of prostatic glands in tissue recombination through maintenance of stem/progenitor cell markers CD44, p63, alpha2beta1 integrin, and CK5.3) Fusion of human prostate epithelial (NHP) cells with tumor cells and inhibition of tumorigenicity in NHP-tumor cell hybrids by p16 and hTERT.
