CurrentAt Novartis I have worked in antibody discovery, with particular focus on yeast and phage display, and high throughput automated assay design and programming,Using yeast display, I improved the potency of an anti-BCMA antibody over 1000x, leading to an IND filing for a CD3 bispecific antibody and a granted patent. I designed and created the maturation library, performed the selection campaign, subcloned and expressed the bispecific antibodies in microplate scale, and ran high throughput activation assays to select the lead molecules.For that project, I developed, optimized, and programmed a variety of automated processes which would later become standard workflows in our group, including a plate based miniprep protocol, 96 well mammalian cell transfection and expression, and a high throughput activation assay. Another useful assay I designed and programmed is a high throughput flow cytometry binding assay, with labeling on a Biomek i7 system and readout using an iQue analyzer. I used this protocol to identify single domain antibody hits for a radioligand therapeutic project. This has become very commonly used in our group as a primary screening method.
