I have led multidisciplinary teams that have brought therapeutic antibodies to the clinic. My extensive hands-on laboratory skills include all aspects of molecular biology from traditional to the latest techniques such as ligation-independent methods (i.e. Gibson assembly).Highlights:At Alder Biopharmaceuticals I was the project leader for the CGRP migraine program from inception of the project through antibody discovery and on to rodent preclinical IND-enabling studies. The resulting clinical candidate, ALD403, has successfully completed several phase II studies and is now in phase III. Personally humanized two antibodies that have been through phase II clinical trials that have shown remarkably low immunogenicity (clazakizumab and ALD403). Overall I have successfully humanized over 50 clinically-relevant antibodies.Performed many of the early experiments and optimization studies for Alder's unique antibody yeast expression platform which has been used for commercial manufacturing in support of clinical trials.Significant experience with antibody engineering including isotype switching, Fc engineering, humanization, bispecific antibody platforms and affinity maturation via phage display.Developed robust molecular biology protocols used for direct cloning of variable domains from single B cells. These protocols are currently being used by several biotech and pharmaceutical companies to support antibody discovery efforts.As the initial employee at Alder, I was instrumental in laying the foundation of what has become one of Seattle's premier biotech companies.
