Postdoctoral Scholar
Worked to understand the mechanisms that drive the development of fat cells in dynamic environments. I designed and built a fluidic platform that enables independently-programmed exchange of signaling hormones, for each of several different populations of developing adipocytes, while simultaneously monitoring these cells using fluorescent microscopy. I used my single-cell imaging analysis tools to measure how synergistic and dynamically-changing hormones work to inhibit or encourage differentiation. I have also established and validated a CRISPR-tagged mouse line that enables single-cell study of adipocyte development in single primary (or potentially in vivo) cells.
Postdoctoral Scholar
Used macrophages derived from a reporter mouse line I designed to measure and understand stimulus-specific NF-κB dynamics in primary bone-marrow macrophages. This work demonstrated that immune cells are capable of reliably encoding different types environmental information (here, the bacterial, viral, or endogenous origin of a molecule) in the oscillations or sustained activity of this transcription factor.
Ph.D Student
I developed an automated live mammalian cell-tracking system for live-cell microscopy, and applied it to measure variable dynamics of an important immune transcription factor, NF-κB, in single macrophages. I applied information-theoretical approaches to demonstrate that these activation dynamics encode non-redundant information about a cell's external conditions. Finally, I coupled these single-cell dynamic studies with traditional knockout studies and computational modeling to understand network mechanisms that shape Toll-like receptor signaling in innate immunity.
