An active member of the In Vitro Disease Modeling (IVDM) Group. Provide immune-oncology and neurobiology project teams with disease and drug mechanism of action (MOA) information through the use of gene expression and functional genomic analysis using a variety of Next Generation Sequencing (NGS) and other methods. Perform bulk RNA/DNA sequencing in-house utilizing the ThermoFisher Ion GeneStudio S5 Prime System. Perform single cell RNAseq including V(D)J TCR/BCR, and CITESeq using the 10X Genomic’s platform to generate libraries in-house while outsourcing the sequencing. Samples range from cell lines, blood, excised tissues, tumors, spheroids, and iPSC derived organoids.

Design and implementation of image based phenotypic small molecule library screens in the neurobiology arena.Screen for small molecule inhibitors of axonal (Wallerian) degenerationDeveloped a high throughput 384 well, label-free, image based assay utilizing dissociated rat dorsal root ganglion (DRG) neurons. Vincristine, a microtubule destabilizer, was used as a chemical injury agent to screen small molecule libraries for inhibitors of axonal degeneration. Responsible for cross-site transfer of the assay and cross-site management of the screen at our Vertex San Diego facility. Developed a low-medium throughput 96 well axotomy (severed axon injury) assay as a confirmatory assay for the original library screen hits and to find compounds which protect under both injury conditions. Active participant in Axon Protection pre-project team with med-chem support; selection of lead compounds and follow-up compound SAR.Screen for small molecules that induce oligodendrocyte differentiation and/or enhance myelin/myelinationImplemented 96 and 384 well, image based screens utilizing P1 rat mixed primary cortical cultures. Hits from the initial screen are further assessed for their ability to myelinate the neurons in longer duration P1 rat mixed primary cortical cultures.

I worked as part of a small imaging core group utilizing The BD Pathway imaging system capable of both standard fluorescence and confocal imaging. My responsibilities include developing low to medium throughput 96 well imaging assays that demonstrate small molecule target specific responses (Mechanism Based Cellular Assays, MBCA), and generating dose response curves. The imaging assays I have designed to date are a mix of live and fixed cell utilizing both standard and confocal imaging techniques.
