Design and implementation of image based phenotypic small molecule library screens in the neurobiology arena.Screen for small molecule inhibitors of axonal (Wallerian) degenerationDeveloped a high throughput 384 well, label-free, image based assay utilizing dissociated rat dorsal root ganglion (DRG) neurons. Vincristine, a microtubule destabilizer, was used as a chemical injury agent to screen small molecule libraries for inhibitors of axonal degeneration. Responsible for cross-site transfer of the assay and cross-site management of the screen at our Vertex San Diego facility. Developed a low-medium throughput 96 well axotomy (severed axon injury) assay as a confirmatory assay for the original library screen hits and to find compounds which protect under both injury conditions. Active participant in Axon Protection pre-project team with med-chem support; selection of lead compounds and follow-up compound SAR.Screen for small molecules that induce oligodendrocyte differentiation and/or enhance myelin/myelinationImplemented 96 and 384 well, image based screens utilizing P1 rat mixed primary cortical cultures. Hits from the initial screen are further assessed for their ability to myelinate the neurons in longer duration P1 rat mixed primary cortical cultures.
