About Bruna Petry
Postdoctoral Research Associate at Iowa State University. Background in molecular biology, animal biotechnology and bioinformatics. My expertise includes wet-lab techniques as PCR, qPCR, cell culture, DNA and RNA extraction, siRNA, gene editing (CRISPR/Cas9), and Illumina libraries protocols. Additionally, I am skilled in utilizing bioinformatic tools for RNA sequencing and allele-specific analysis.During the COVID-19 pandemic, I actively contributed to diagnostic efforts through qPCR testing and the sequencing of new SARS-CoV-2 variants using Illumina libraries (Illumina COVIDSeq).

Postdoctoral Fellow
Postdoctoral Research Associate (Bioinformatics)
Leading different projects from Functional Annotation of Animal Genome (FAANG) project involving cattle and pig species: i) allele-specific epigenetic analysis; ii) development of a web tool for omics data analysis linking genotype to phenotype; iii) exploration of alternative-splicing phenomena. iv) working with RNA-seq pipeline to develop gene expression atlas for cattle and pig.v) engaged in collaborative efforts with both national and international research groups, particularly contributing to projects centered around allele-specific analysis in pigs and cattle.
Graduate Student, Doctorate (Cell And Molecular Biology)
Project Title: Evaluation of a Gene Associated with Muscle Development in Chickens through CRISPR/Cas9 Gene EditingProject Overview:i) Poultry farming has seen substantial genetic advancements, boosting production and reducing costs.ii) New methodologies aim to identify genes and genetic variations for economic traits in poultry.Challenges remain in accurately establishing genetic changes' effects on the phenotype.iii) Utilizing CRISPR/Cas9 genome editing to knockout or edit genes and validate the effects on the phenotype.Advancing our understanding of how genetic changes regulate phenotypic characteristics in poultry.iv) The objective was to identify candidate genes or mutations for regulating muscle mass deposition in broiler chicken lines. Test the effect of the identified gene through gene editing in muscle cell cultures. Contributing to cutting-edge research at the intersection of genetics and poultry production.v) Exploring the potential of gene editing and siRNA to enhance poultry traits.

Project Title: Functional Analysis of the SAP30 Gene and Its Relation to Muscle DevelopmentProject Overview:i) Chickens are a vital protein source, with muscle growth being a key trait in poultry breeding programs.ii) Genetic advancements have enhanced poultry yield and efficiency, reducing production costs.iii) Understanding the molecular processes behind muscle development is crucial for identifying responsible genes.iv) New methodologies are being used to identify candidate genes and genetic variations linked to muscle traits. However, few causal genetic variants associated with phenotypes have been uncovered.v) Genome editing techniques like CRISPR/Cas9 and small interfering RNAs (e.g., siRNA) hold promise for validating or identifying genetic effects on phenotypes.vi) For that we Investigated and validated the role of the SAP30 gene in muscle development, using RNA sequencing to identify differential gene expression and co-expression patterns and gene networks in relation to SAP30 treatment.vii) The results provide experimental evidence of the involvement of SAP30 gene as a regulator of muscle cell hypertrophy
Graduate Student, Master (Poultry Genetics)
Project Title: Validation of Differentially Expressed Genes in Broiler Chickens' Femur Transcriptome with and without Femoral Head NecrosisProject Overview:i) Bone health issues impact the poultry industry, causing economic losses, reduced performance, and compromised animal welfare.ii) Bacterial chondronecrosis with osteomyelitis (BCO) is a common bone anomaly in broiler chickens due to rapid genetic growth.iii) Our objective is to validate genes linked to BCO in broiler chickens, utilizing 10 healthy and 10 BCO-affected birds.iv) The transcriptome data was previously generated at Embrapa Swine and Poultry using total RNA sequencing (RNAseq) on the femur's growth plate in Cobb 500 lineage male commercial birds.v) As results Differentially expressed (DE) genes between normal and BCO-affected broilers were found. ADIPOQ, ANGPTL5, SFRP5, COL14A1, ANGPTL7 and COL8A1 are strong candidate genes related to BCO. The DE genes evaluated contribute to the development of BCO in chickens.
Bachelor In Biological Science (Internship In Poultry Genetics)
Title: Prospecting for Genes Associated with Bone Integrity in ChickensHighlights of my two-year internship during my undergraduate studies:i) Conducted sample collection from poultry and one-day-old chickens.ii) Managed, stored, and processed collected samples.iii) Gained expertise in DNA extraction, PCR, and genotype analysis using the RFLP technique.iv) Proficiently performed RNA extraction from poultry muscle and bone tissues.v) Contributed to research focusing on investigating associations between candidate genes and production traits in the poultry industry.Leveraged my internship to develop hands-on skills in genetics and contribute to valuable poultry research.
