About Bryan Ferguson
I’m an early-career scientist training in drug development for neurodegenerative disorders based on novel biological mechanisms driving disease progression. I have a specific focus on neuroimmunology and microglial biology with experience studying Alzheimer’s Disease and progressive multiple sclerosis. I have worked as a research associate in the lab of Stuart Schreiber at the Broad Institute of MIT and Harvard investigating mechanisms of neurodegeneration and the role apolipoprotein E (apoE) plays in regulating the lipidome and redox balance in CNS cell types. Working largely independently, my work has provided insights into mechanisms by which apoE isoforms confer vulnerability to or protection against neurodegenerative processes involved in late-onset Alzheimer's disease and other neurodegenerative disorders. These insights include isoform-dependent differences in lipoprotein trafficking through various organelles, isoform effects on lipid debris clearance in glia, and isoform effects on glial sensitivity to ferroptosis. In order to gain specialized exposure to neuroimmunology, I then work at PureTech Health where I worked as an Associate Scientist on a team studying the effects of aging on meningeal lymphatic function. I led the efforts developing sample workflows for bulk RNA-seq and scRNA-seq rodent aging studies, developing protocols for tissue homogenization, RNA-extraction and purification, cDNA library synthesis, and single cell preparation and characterization. I currently work as a Principal Associate Scientist in the Multiple Sclerosis and Immunology Research Unit at Biogen, working on both target discovery and exploratory biology as well as late-stage preclinical drug discovery programs.

I worked on a team investigating the effect of age and neuroinflammation on meningeal lymphatic function. I worked to develop bulk RNA-seq and scRNA-seq sample workflows from mice and rats in order to generate high dimensional datasets capable of informing target identification and therapeutic development. I also helped develop IHC protocols to study meningeal T cell localization and VE-Cadherin endothelial cell-cell junctions. Finally, I trained to carry out rodent survival surgeries for an in vivo imaging assay measuring CSF drainage to the deep cervical lymph nodes (dCLNs)

I work in the Schreiber Lab in the Chemical Biology and Therapeutics Science Program investigating the potential applications of ApoE lipoprotein and apoE-related biology as a novel therapeutic target to treat various forms of neurodegeneration, including late onset Alzheimer’s disease (LOAD). My work has helped elucidate important differences in isoform-dependent differences in lipoprotein trafficking, which constitute novel and potentially important insights into how apoE isoforms either confer protection or risk for the development of LOAD. I have also helped lead studies demonstrating sensitivity of microglia and astrocytes to ferroptosis, modulation of ferroptosis sensitivity by mciroglial state polarization and small molecule treatment, as well as phagocytosis of lipid debris.

I served as the committee co-chair of Out@Broad, the Broad Institute’s LGBTQIA+ employee resource group. I help lead transformative efforts to foster a more inclusive environment at the Broad in order to empower LGBTQIA+ members of the scientific community. These efforts include but are not limited to developing a comprehensive policy to create universal gender-inclusive bathrooms in order to foster a more gender affirming environment at the Broad.
