I both manage an individual project and contribute to a research team team tasked with optimizing the design and construction of engineered nucleases for targeted therapy in the human genome. Research efforts focus on the employment of nucleases and nickases for the repair of the genetic mutations that are the underlying causes of sickle-cell anemia and cystic fibrosis. Nucleases employed include transcription activator-like effector nucleases (TALENs), zinc finger nucleases (ZFNs), and clustered regularly interspaced palindromic repeats (CRISPRs). A second research emphasis centers around the optimization of DNA repair pathway choice. We aim to design nucleases that induce repair preferentially by either homologous recombination or non-homologous end joining. Additional efforts focus on more efficient delivery methods of engineered nucleases.
