PhD in molecular virology studying classical swine fever (CSFV), a pestivirus belonging to the Flaviviridae family. I worked with optimization of full-length cloning (12.3 kb) of RNA viruses, using In-Fusion and TOPO-XL2 cloning, in order to haplotype and map virus subpopulations. The E2 glycoprotein of CSFV is responsible for the viral binding to host cells, and my focus has been on a specific motif herein (aa 761-764). Mutants were created by site-directed mutagenesis and tested in vivo in pigs. Tissues from pigs were analyzed by NGS to reveal differences in quasispecies composition . Mutants were also tested in vitro in different cell lines to reveal their role in cell specificity. Show less
