About Carla Morales Ferré

He trabajado como investigadora postdoctoral en el grupo de investigación Inmunología de la Malaria en ISGlobal donde he participado en el estudio longitudinal de seguimiento sobre SARS-CoV-2, en el cual evaluamos la respuesta inmunitaria de los linfocitos T en el contexto de exposición natural y vacunación. Previamente, en 2018 terminé el trabajo de fin de máster en el cual desarrollmos células dendríticas tolerogénicas para tratar ratones con encefalomielitis autoinmune experimental (modelo animal de esclerosis múltiple). En 2022, finalicé mi doctorado en la Universidad de Barcelona en el cual estuve realizando diferentes estudios preclínicos relacionados con el desarrollo del sistema inmunitario neonatal. Evaluamos el papel del dimorfismo sexual y del efecto de dos suplementos dietéticos en el desarrollo del sistema inmunitario y su rol para prevenir y aliviar la infección causada por Rotavirus durante la lactancia. Además de la experiencia adquirida en la realización de diferentes técnicas experimentales, he elaborado informes de proyectos, publicaciones, protocolos, planes estadísticos y he realizado búsquedas bibliográficas exhaustivas. Asímismo he participado en la organización de eventos científicos, en la preparación de material científico para conferencias y he realizado comunicaciones orales. Durante mi carrera investigadora siempre me he centrado en el enfoque traslacional de la ciencia, por ese motivo he cursado el Máster en Monitorización de Ensayos Clínicos y Medical Affairs donde actualmente estoy realizando prácticas en Menarini como Medical affairs en el área terapéutica de patologías respiratorias y alérgia.

Título de la tesis: Impacte d'una sobrecàrrega bacteriana i vírica sobre el sistema immunitari de rates lactants i paper preventiu d'un postbiòtic en la diarrea induïda per rotavirusResumen:El sistema inmunitario del recién nacido es inmaduro y, por tanto, predispone al neonato a sufrir infecciones, que en el caso de los recién nacidos del género masculino parecen presentar una incidencia y gravedad mayores que el femenino. La leche materna contiene distintos componentes bioactivos que promueven el desarrollo inmunitario o protegen al recién nacido durante los primeros meses de vida. Las fórmulas infantiles son la alternativa de los recién nacidos que no reciben lactancia materna. Tienen una composición que busca reproducir la de la leche materna, por lo que es frecuente la adición de compuestos bioactivos como los prebióticos, probióticos y posbióticos.Objetivo: establecer los efectos sobre el desarrollo del sistema inmunitario neonatal de dos infecciones comunes durante el período de lactancia (mastitis infecciosa materna y gastroenteritis vírica del niño) y evaluar el papel del dimorfismo sexual y de los prebióticos scGOS-lcFOS y del posbiótico Lactofidus ® sobre el desarrollo del sistema inmunitario y la infección por rotavirus durante la lactancia.Conclusiones: el contacto con patógenos durante el período de lactancia afecta al desarrollo del sistema inmunitario. Además, el dimorfismo sexual de la respuesta defensiva frente a infecciones parece estar presente en estas primeras etapas de vida. Las intervenciones experimentales de prebióticos y/o posbióticos, parecen una buena estrategia para mejorar el desarrollo microbiano e inmunitario de los lactantes y su capacidad defensiva frente a infecciones, sugiriendo la utilidad de su adición a fórmulas infantiles.

Prácticas Del Máster En Investigación Biomédica Translacional

Trabajo de fin de máster: Combined therapy using antigen-specific tolerogenic dendritic cells and interferon-β in experimental autoimmune encephalomyelitis.Realizado en el grupo de Inmunopatología. Abstract:Multiple sclerosis (MS) is a progressive neuroinflammatory autoimmune disease that cause demyelination and axonal loss, which result in neurocognitive and motor deficits.This study aimed to evaluate the effect of interferon interferon-β (IFN-β), one of the first-line treatments for MS, in combination with antigen-specific TolDC therapy in EAE model.In order to achieve this purpose, EAE was induced to females C57BL/6J mice, 8-10 weeks old using oligodendrocyte glycoprotein (MOG 40-55 ). Mice were treated with TolDC tolerized with vitamin D3 and pulsed with MOG 40-55 (TolDC-MOG), IFN-β or the combination of both treatments (IFN-β + TolDC-MOG). IFN-β was injected subcutaneously from the day 10 post immunization (p.i) to the day 17 p.i and TolDC were injected intravenously on days 14, 18 and 22 p.i. Mice were weighed andexamined daily for clinical signs. On day 29, mice were sacrificed and spleens were obtained to measure the reactivity of splenocytes to MOG 40-55 .Despite no differences were observed in the severity of the disease (maximum and cumulative score) between groups, IFN-β + TolDC-MOG-treated group showed reduced clinical signs compared to monotherapies. In addition, splenocytes from TolDC and IFN-β + TolDC-treated groups showed less auto-reactivity of T cells to MOG 40-55 .In conclusion, a higher amelioration of EAE course was observed in combined-treated group compared to monotherapies with IFN-β or TolDC-MOG. In addition, TolDC-MOG and IFN-β + TolDC-treated groups showed a less auto-reactive splenocytes against MOG 40-55 . These results could indicate that IFN-β can increase the potential effect of antigen-specific TolDC in EAE and in MS patients.

Título del trabajo de final de grado: Immunomodulation of probiotics in pregnant rats.Realizado en el grupo de Autoinmunitat i Tolerància de la Facultat de Farmàcia i Ciències de la Alimentació.Abstract:In the recent years, it has been a significant increase in research on the characterization and verification of the potential health benefits associated with the use of probiotics. This study aimed to investigate the safety of supplementation with a probiotic from the genus Lactobacillus and its effect on the immune system.In order to achieve this purpose, female adult Wistar rats were orally supplemented with the probiotic from the beginning of gestation to the 14th day of lactation. During the gestation and lactation periods, rats were routinely weight and faeces were obtained by an abdominal massage in order to observe changes in the body weight or faecal consistency. On day 14th of lactation rats were sacrificed. Rats’ weight and length were measured, organs were weight and intestinal lavages, mammary glands, submaxillary glands and mesenteric lymph nodes were obtained in order to quantify the levels of IgA and IgM by ELISA.The probiotic supplementation did not influence neither the weight of rats nor the weight of organs, the body mass index and Lee index were similar in both groups. The consistency of the faeces did not change between groups and the levels of IgA and IgM in the intestinal compartment as well as in the other extraintestinal mucosa were not modified. However, a decrease in faecal pH between the days of lactation 7th and 14th was observed.In conclusion, the supplementation with the strain of Lactobacillus in pregnant rats is safe and does not modulate the production of the two immunoglobulins (IgA and IgM) assayed. Moreover, the decrease in pH is observed at the end of the lactation period could indicate the colonization of the probiotic
