About Caroline Stefani

My research focuses on understanding the cellular response to damage and signaling induced by pathogen. I particularly focus on bacterial toxins and their effects on endosomal trafficking and actin cytoskeleton remodeling using microscopy, biochemistry, and genetic approaches. My early work clarifies the cytotoxic effects induced by toxins of B. anthracis on the actin cytoskeleton of primary endothelial cells. These studies allow us to correlate the actin remodeling and the cell plasticity to vascular leakage. Moreover, we found that the induction of transcellular tunnels is triggered by a physical phenomenon, newly called cellular dewetting, that we could model. Our discovery, that cells damaged by ‘macroapertures’ inducing-toxins initiate a cell-autonomous mechanism of repair, suggests a truly fascinating paradigm. An important part of our host defense against microbes occurs at a cellular level and involves not only the sensing of damage but also the initialization of repair responses. By a novel unbiased forward genetic screen, I currently focus on the characterization of cellular responses to families of bacterial toxins and effectors. To study those intracellular changes, I helped pioneered the development of 3D visualization tool using Virtual Reality (VR). Part of my research also focuses on High Content Imaging (HCI) and the development of automatic image analysis pipelines.
