My two main projects in Dr. Peter Black's lab were screening for small-molecule drug (SMD) inhibitors against Notch2, and evaluating the role of BRN2 in neuroendocrine bladder cancer (NEBC).𝗙𝗶𝗻𝗱𝗶𝗻𝗴 𝗮 𝗡𝗼𝘁𝗰𝗵𝟮 𝗜𝗻𝗵𝗶𝗯𝗶𝘁𝗼𝗿: Dr. Black's group was the first to identify Notch2's role as an oncogene, contrary to the tumour suppressive properties of Notch1. • My role in Dr. Don Moerman's lab was to assist in the production of C. elegans genetic knockout mutants using CRISPR/Cas9. This involved:
• Preparing gene-specific dual-selection cassettes using Gibson Assembly
• Injecting mixes of gRNA, selection cassettes, and Cas9 into the gonads of worms
• Screening for mutants using fluorescence microscopy
