Senior Scientist I - Antibody Discovery
Scientist Iii- Antibody Generation (Phage Display)
- Member of the phage display team in the Antibody Generation group, using naïve and immune-biased phage display libraries to generate large hit sets of antibodies against a wide variety of targets, for use in both.
- Served as Antibody Generation team representative for four project teams. Involved in strategy and decision making, data analysis and presentation, hit selection, and coordination between functional areas (kinetics.
- Panned phage display libraries against protein and carbohydrate antigens, in bead and solid support formats.
- Ran high throughput screening on thousands of phage clones to identify hundreds of unique binders per antibody generation campaign.
- Isolated RNA from immunized mouse splenocytes and lymphocytes, recovered V-genes and constructed an immune-biased phage display library.
- Constructed LC-shuffling and Tumbler phage display affinity maturation libraries in scFv and Fab format.
Research Associate
- Responsible for isolating diverse bacteria from human and mouse intestinal samples and creating large culture collections of a purity necessary for colonization of gnotobiotic mice with defined bacterial communities.
- Processed samples through all steps of a 16s rRNA sequencing pipeline, including sample collection and DNA isolation, PCR, library preparation, Illumina MiSeq sequencing, and data analysis using QIIME2.
- Helped set up and modify a 16s rRNA sequencing bioinformatic pipeline, adapting it to different needs.
- Panned a nanobody phage display nanobody library against a bacterial protein and then isolated, screened, and sequenced individual phage clones.
- Transfected Expi293 cells with vectors expressing each of the 96 chosen nanobody sequences from above and purified the resulting nanobodies.
- Carried out gnotobiotic mouse experiments from beginning to end, involving initial setup and oral gavage, husbandry, ongoing sample and data collection, final dissection, bacterial 16s rRNA sequencing, and ELISA-based.
Undergraduate/Graduate Research Assistant
- Investigated the effects of arginine metabolism on botulinum neurotoxin expression using anaerobic culturing, western blotting, immunoprecipitation, mouse assays, and ClosTron-mediated mutagenesis.
- Involved in the creation of a modified version of ClosTron, a group II intron-based gene knockout method.
- Involved in the study of the conjugation mechanism of a Clostridial plasmid.
- Followed safety, security, and documentation protocols necessary for working independently with Clostridium botulinum and botulinum neurotoxin in BSL-2 and BSL-3 facilities.
- Utilized molecular and microbiological methods including immunoprecipitation, western blotting, cloning, PCR, RNA and DNA isolation, ClosTron gene inactivation, anaerobic culturing, and bacterial conjugation.
Undergraduate Research Assistant
- Studied the population genetics of Mycobacterium tuberculosis.
- Participated in the building of a bioinformatics pipeline.
- Processed and analyzed whole genome sequencing data and wrote scripts to automate and streamline data processing.
