Profile bio

About Chi (Aimee) Zhao, Ph.D.

The primary efforts of my Ph.D. research are directed towards engineering chimeric transmembrane proteins for both translational and fundamental studies. Specifically, the goal of my translational research is to develop cell-derived biomaterials for specific and efficient drug delivery. To fulfill this aim, I focused on 1) engineering multifunctional transmembrane proteins for targeting cell-derived vesicles to cancer cells, and 2) decorating these vesicles with high concentrations of transmembrane channel proteins to facilitate the unloading of macromolecular drugs directly into the cytoplasm, maximizing their efficacy. Using the same chimeric proteins developed in these two projects, my research in fundamental science focuses on understanding the underlying mechanism of receptor uptake by endocytosis, an essential pathway for drug delivery vehicles to enter cells.Working as an experimentalist, I have 5+ years of wet lab experience with proficiency in mammalian cell culture, fluorescence microscopy, flow cytometry, molecular cloning, lentiviral transfection, gene editing, and protein purification. Working as a data analyst, I have performed complex analyses to microscopy data collected from mammalian cells using MATLAB. Further, I have 5+ years of experience in using Adobe Photoshop and Illustrator to create illustrations for my publications. My original research so far has been published as 7 first-authored or co-authored peer-reviewed papers with 3 more to come. During my Ph.D., I have mentored 8 undergraduate researchers, and 2 junior graduate researchers. My experience in being a team leader has taught me how to adapt to and foster a collaborative environment. I am looking to leverage my interdisciplinary background in applied and fundamental science to work as a R&D scientist in biotechnology or pharmaceutical companies. As a highly motivated individual, my goal is to make my own contribution in pushing the current therapeutic agents to the next level.

Listed skills include Molecular Biology, Data Analysis, Microsoft Office, Flow Cytometry, and 25 others.

1. Developing cell-derived biomaterials for specific drug delivery- Designed multifunctional transmembrane proteins that target cells on the basis of their surface receptor expression profiles. Further enhanced targeting selectivity by including serial repeats of the targeting ligand domain on the chimeric transmembrane proteins.- Genetically engineered donor cells to stably express multifunctional transmembrane proteins and harvested their plasma membranes as drug delivery vehicles.Cover photo: http://onlinelibrary.wiley.com/doi/10.1002/smll.201670136/fullPublication: http://onlinelibrary.wiley.com/doi/10.1002/smll.201600493/fullhttps://pubs.acs.org/doi/abs/10.1021/acs.biochem.7b006882. Engineering cell-derived biomaterials for efficient macromolecular delivery directly to the cytoplasm- Genetically engineered donor cells to stably express transmembrane channel proteins, connexin 43, providing macromolecules with a direct passageway into the cytoplasm- Designed and expressed a chimera version of connexin 43 protein with a RNA binding domain taken from virus in order to increase the concentration of siRNA in the cell-derived plasma membrane vesicles to enhance gene silencing- Quantitatively measure the copy number of siRNA in donor cells and cell-derived vesicles using qPCR3. Investigating the impact of receptor heterodimerization on the receptor uptake mechanism by endocytosis- Designed and constructed chimeric transmembrane model receptors in order to establish a simplified model system in live cells for this study- Used quantitative confocal imaging as well as publicly available MATLAB packages to map the distribution of receptors on the plasma membrane of cellsPublication: https://www.sciencedirect.com/science/article/pii/S0006349519305685https://pubs.rsc.org/am/content/articlelanding/2019/sm/c9sm00876d
