    Under the Supervision of Robert Lu Performed transfection of PER.C6® cells with DNA plasmids containing recombinant protein sequences for the production of biotherapeutics. PER.C6® stable pools are generated post transfection via culturing in selective media and are used to perform limited dilution cloning. Following limited dilution cloning, 300-500 clones are generated . Clones are titered for protein expression levels and both the protein product and clones are characterized. Clones with profiles for growth, protein production, stability and product quality that do not meet predetermined criteria are discarded until a final clone is selected. Duties include DNA preparation, cell culture, transfection of PER.C6 cells, stable pool generation, limited dilution cloning, batch and fed-batch assessments, titer assessments, cell banking and presentations.

    Under the Supervision of Martin Stone.Thesis Title: Towards a model of chemokine/receptor complexes: complementation of chimeric receptors and mutant chemokines To determine which extracellular regions of the chemokine receptor CCR3 were important for specificity and activation, the extracellular regions of CCR3 were replaced with those of the related chemokine receptor CCR2. Fifteen receptor mutants were cloned from the cDNA, using recursive PCR, encompassing all possible combinations of extracellular regions. Several mammalian cell lines were transfected with the receptor cDNA constructs and cultured in selective media. Receptor constructs were tested for changes in specificity and activation through the use of mutant and wild type chemokines in conjunction with radioligand binding and calcium flux assays.
