About Chris Buhr

I am a medicinal chemist that either directed or helped drive the discovery of multiple NCE's that led to the filing of IND applications and Clinical Trials. I have a proven track record of success in the PI3K and mTOR arenas, as well as expertise with the inhibition of other kinases and targets, including cKit, Chk1, Jnk, and Hsp90. I have extensive experience with the modulation of nuclear hormone receptors including RORgamma, FXR and LXR. I have approximately forty total patents and applications twenty U.S. Patents.ACCOMPLISHMENTS: CLINICAL TRIALS AND INDsI either led, or helped drive, six projects to IND filings. Five of the new chemical entities entered human clinical trials, mostly in oncology. Eleven Phase I and Phase II clinical trials were run on voxtalisib.• CC-401: JNK Inhibitor (IND, Phase I, Signal / Celgene)• XL844: Chk1 Inhibitor (IND, Phase I, Partnered with GSK)• XL335: FXR Agonist (IND, Phase I, Partnered with Wyeth as WAY-362450)• XL765: PI3K/ mTOR Inhibitor (IND, Phase II, Voxtalisib, Partnered with Sanofi)• XL888: HSP90 Inhibitor (IND, Phase I)• XL388: mTOR Inhibitor (IND Filed)

Chris Buhr Work Experience Details
  • Visionary Pharmaceuticals
    Consultant
Visionary Pharmaceuticals Apr 2020 - Present
    San Diego, California, United States
  • Trisaq, Inc.
    Vice President Of Chemistry
    Trisaq, Inc. Oct 2019 - Apr 2020
    Cambridge, Ma
I led a program to make allosteric agonists of human dynamin to treat Chronic Kidney Disease (CKD). Dynamin modulates actin and the cytoskeleton involved in maintaining foot process integrity in podocytes. Loss of foot process integrity in podocytes gives rise to proteinuria and CKD.
  • Visionary Pharmaceuticals
    Vice President Of Medicinal Chemistry
    Visionary Pharmaceuticals Apr 2018 - Sep 2019
    San Diego, Ca
I directed all medicinal chemistry efforts on the antagonism of RORgamma, an important modulator of IL-17.
  • Visionary Pharmaceuticals
    Senior Director Of Medicinal Chemistry
    Visionary Pharmaceuticals 2013 - Mar 2018
    San Diego, Ca
I directed the medicinal chemistry efforts on a RORgamma modulator program. RORgamma is a nuclear hormone receptor that is implicated in a number of inflammatory diseases, alcoholic liver disease, and NASH. I directed all of the chemistry, and many supporting biological studies, through contract research organizations.
  • Consultant
    Pharmaceutical Consultant
    Consultant 2012 - 2013
I provided drug discovery consulting services to the pharmaceutical industry.
  • Exelixis
    Director Of Medicinal Chemistry
    Exelixis 2006 - 2011
    South San Francisco, Ca
I directed major research and development projects, from conception to clinical trial and/or IND filing. Orchestrated key cross-functional partnerships to support project success. Led drug discovery initiatives and delivered final products for transfer to process chemistry.Highlights• Drove all aspects of medicinal chemistry efforts for developmental of XL765 PI3K/mTOR inhibitor, voxtalisib, for solid tumor cancer treatment with potential for large market. The inhibitor was in eleven Phase I and Phase II clinical trials with partner Sanofi Aventis as SAR245409. Voxtalisib was in clinical trials for eleven years.• Coordinated and co-directed medicinal chemistry efforts in development and IND filing of selective mTOR inhibitor (XL388).• Provided key support in discovery of HSP90 Inhibitor (XL888) that entered clinical trials.
  • Exelixis
    Associate Director Of Medicinal Chemistry
    Exelixis 2002 - 2006
    South San Francisco, Ca
Led a team of medicinal chemists in a wide range of projects, focussing on metabolism and oncology targets. Moved key projects to advanced research stages. Supported efforts of additional research projects and focuses. Led larger teams of medicinal chemists (over 15) as project leader.Highlights• Directed medicinal chemistry efforts to advance Chk1 protein kinase inhibitor (XL844) to human Phase 1 clinical trials for cancer. XL844 was first small molecule Chk1 inhibitor to enter clinical trials.
  • Exelixis
    Senior Research Scientist
    Exelixis 2001 - 2002
    South San Francisco
Managed all medicinal chemistry aspects of Chk1 inhibitor project. Supervised large chemistry team as the chemistry project leader.Highlights• Drove significant improvements in the in vitro and cellular potencies of Chk1 inhibitors, resulting in filing of patent application and advancement of project to preclinical stage.
  • Signal Pharmaceuticals / Celgene Corporation
    Senior Research Scientist
    Signal Pharmaceuticals / Celgene Corporation 1999 - 2001
    San Diego, Ca
Synthesized selective low nanomolar Jun kinase (JNK) inhibitors. Studied in vitro, cellular, and animal structure activity relationships, as well as pharmacokinetics of these molecules. Highlights• Synthesized and discovered CC-401, which entered Phase 1 clinical trials.
  • Gilead Sciences
    Research Scientist
    Gilead Sciences Jun 1988 - Jul 1999
    Foster City, Ca
I was the sixth hire, and second Ph.D. at Gilead Sciences. Led various research projects, with special focus on synthesis and structure-activity relationships of small antiviral molecules, nucleosides, and oligonucleotides. Synthesized numerous heterocycles, nucleosides, oligonucleotides, and antiviral compounds.
