Project 1: Developed an ex vivo imaging assay to investigate gender-specific regulation of bulge stem cell proliferation in a murine model• Trailblazed and optimized conditions to maximize tissue survival and subsequent pharmaceutical compound screens to monitor changes in stem cell proliferation and/or migration.• Targeting HSP90 using mouse and human cell cultures, using tissue isolation and MACS cell sorting to enrich for stem cells with subsequent analysis by qPCR, IHC/IF, Chromatin Immunoprecipitation (ChIP).• Followed UCSD ACP guidelines and protocols for humane treatment of animals during mouse handling, breeding, genotyping, tissue dissection, and surgical procedures.Project 2: Novel epidermal differentiation and enhancer function using an activated BRAFV600E model• Analysis of p300 binding sites in skin from p300 ChIP-Seq data.• Target identification and cloning of 60+ enhancer elements bound by EP300 via gateway cloning.• Produced, isolated, and quantified lentivirus constructs followed by transfection of NHEKs.• Analysis of transfected cells through Fluorescent microscopy, Time-lapse microscopy, IHC/IF, and qPCR.• Utilized an in-house high throughput mouse model of Xeno-transplantation for screening, characterization, and analysis of previously identified enhancer constructs.
