Special course & Master Thesis in the Mammalian Cell Line and Bioprocess Engineering Group. The project's aim was to study in depth the role of signal peptides in the secretion pathway in Chinese Hamster Ovary cells. The productivity of three monoclonal antibodies was evaluated from the screening of different signal peptide combinations. For that, I designed and performed a high-throughput cloning experiment to generate a large set of plasmid vectors. I later evaluated the cell productivity in a high-throughput transient transfection experiment in CHO cells. From the results, relevant features of both signal peptides and antibodies were analyzed. Skills acquired: design of high-throughput experiments, optimization of cloning methods, skilled in different cloning techniques (USER cloning, NEBuilder, PCR mutagenesis) for vector construction, restriction enzyme digestion, media preparation, bacterial culture, DNA extraction methods, whole plasmid sequencingMammalian cell culture: sterile technique, cell counting, cell imaging (Celligo), transfection, transient evaluation, flow cytometry and FACS sorting, liquid handler experience (E2P2), measurement of antibody production with Octet biosensor, generation of stable cell lines through RMCE, junction PCR, qPCR.Statistical analysis: JMP, Minitab, Excel, Graphpad Prism.Literature review and presentation of results.
