- Conduct experiments and analyses regarding protein-DNA interactions of homeodomain proteins focusing on the Paired-like family and its ability to be cooperative or not on AT rich motifs - Responsible for cloning over 60% of all Paired-like factors found in humans and perform Native Protein Purification as well - Utilize EMSAs and Luciferase Reporter Assays for protein characterization specifically looking at cooperativity, affinity and specificity of Paired-like factor Wildtype and Disease Variant proteins found in humans- Use Drosophila Melanogaster to study human disease variants of Paired-like factors and how they affect function and gene expression in-vivo with the goal of rescuing the fly- Collaborate with other labs at CCHMC and UC Medical Center to improve current experiments and discuss projects- Present data and future experiments at lab meetings to gather insight and discuss methodology for potential grants
