Postdoctoral Researcher
Postdoctoral researcher in Charles Melnyk groupPlant Biology Department - SLUPlant grafting is an ancestral technic, consisting of joining two plants to create one chimeric plant. However, the molecular mechanisms of grafting capacities in plants remain poorly understood. In this context, my research project aims at identifying genes involved in graft formation in plants, using Arabidopsis thaliana and tomato plant models.Main achievements:- Generation of transgenic material in Arabidopsis thaliana and tomato- In-depth phenotyping characterization of mutant lines, presenting defects in regeneration- Protocol optimization for micrografting of Solanaceae species, and in different temperature conditions. This work was part of a project published in Development in 2022.- Protocol optimization for monocots grafting, in collaboration with the University of Cambridge. This work led to a publication in Nature in 2021.
Doctoral Student
Fruit tissue morphology is a key character defining the final fruit quality. However, the cellular and molecular mechanisms by which fruit tissue morphology is defined remain incomplete. In this context, the aim of my thesis was to identify and characterize new regulators of fruit tissue morphology. Through a forward genetic strategy, I investigated the role of a Guanylate Binding Protein (GBP) protein at the origin of a thin pericarp phenotype in tomatoes. I studied in parallel the role of the protein in tomato and Arabidopsis thaliana models. My work suggests a role for the GBP in the fine control of cell division in the tomato pericarp, and was published in Plant Cell in 2020.Main achievements:- Mapping-by-sequencing strategy (according to Garcia et al., 2016)- DNAseq analyses- In-depth phenotyping characterization of tomato EMS mutant- Generation of Arabidopsis thaliana mutant lines using CRISPR-Cas9- Generation and maintenance of transgenic BY-2 tobacco cells- Tissue preparation for RMN and LC-MS analyses

Phenotypic characterization (at the fruit, tissue, cellular and molecular levels) of tomato EMS mutants presenting alterations in fruit growth and endoreduplication.
