Bachelor's Thesis: Role of the P2X7 receptor in ozone-induced airway hyperresponsiveness and airway inflammationABSTRACT: Preliminary data has shown that ozone irritates epithelial cells resulting in the release of adenosine triphosphate (ATP). The P2X7 protein (P2X7) is a purinoceptor ligand-gated ion channel found on mast cells and is involved in extracellular signaling by ATP, a well-recognized pro-inflammatory factor in a number of diseases including asthma. To test the impact of the P2X7 purinoceptor in asthma exacerbations, airway hyperresponsiveness (AHR) to methacholine (MCh) and airway inflammation was measured in P2X7 knockout (KO) mice and wildtype (WT) mice after exposure to ozone. AHR following ozone exposure was significantly attenuated in P2X7 KO mice, while airway inflammation showed no difference from WT control mice exposed to ozone. These results demonstrating a critical role for the P2X7 receptor in the development of ozone-provoked AHR suggests that this receptor may represent a novel therapeutic target to reduce the impact of asthma exacerbations by this common environmental pollutant.
