I have a strong background in production, purification and structure determination of a-helical membrane proteins by NMR and X-ray crystallography. Specifically, I have been involved in understanding signaling events that take place at the cellular membrane by investigating sulfurtransferases, potassium channels and the essential malarial protein-conducting pore complex. In addition, I contributed to rapid NMR structure determination of human integral membrane proteins produced via cell-free expression systems. As alternative membrane mimetics, I described in detail the structure of lipid nanodiscs by NMR, used them to reconstitute membrane proteins, and characterized membrane proteins in nanodiscs by label-free single molecule mass photometry. During this time, I also got familiarized with cryo-EM to investigate structures of amyloid fibrils and combined advanced biophysical and cell biology approaches to better understand the functions of the Parkinson’s disease protein a-Synuclein.
