Insecticidal protein discovery and analysis.Senior Research Associate (SRA) III (2011 – 2018), SRA II (2001 – 2011)• FPLC purified insecticidal proteins from plant, bacteria, and fungal sources leading to numerous novel protein discoveries for deployment in transgenic plants.• Used E. coli expression systems and protein purification (analytical to gram scale) for construct characterization and high purity protein production for use in protein characterization, mode of action studies, and regulatory studies.• Prepared trypsin digest samples of PAGE gel bands and chromatography fractions for MS/MS analysis to evaluate lead purification and enable rapid purification progress minimizing time spent on previously identified inhibitory proteins.• Performed PAGE and Western blotting to evaluate transgenic plants for target protein expression enabling rapid transgenic event sorting and correlation of protein expression with plant efficacy data.• Ran simulated gastric fluid digestion and SDS PAGE analysis to evaluate proteins for digestibility and identify potential allergenicity problems. • Achieved production and purification of selenomethionine labeled proteins for X-ray crystal structure studies using defined growth media and a methionine auxotroph expression cell line.
