He developed several techniques to probe the structure of regions of a chromosome in the fruit fly, a widely used model organism. He demonstrated that genes which are turned off, by a mechanism called Polycomb repression, are physically inaccessible to proteins in the nucleus. It took years of persistence to set up the assays, and skillful genetic and molecular work to generate the results. The major publication from his thesis (Fitzgerald and Bender (2001) Mol. Cell Biol. 21:6585-6597) is still the best evidence for chromosome compaction by Polycomb repression. Dan did it all on his own, and by the end of his time, he was more familiar than I with the relevant literature, and more thoughtful about experimental details.
