I'm currently a PhD student at Newcastle University, specialising in the molecular basis of cancer. Specifically, I am interested in early genetic fusion events during prostate tumourigenesis with a strong emphasis on generating clinical models such as novel cell lines and 3D organoids that recapitulate the disease phenotype. Previously, I held a position as a Research Scientist at the Northern Institute for Cancer Research, where I was tightly involved in a multi-national project that acquired daily human tissue samples for processing and analysis. Additionally, I have gained experience in the design and execution of multiple experiments simultaneously, with a strong emphasis on biochemical assays such as BCA/ ELISA/ Luciferase assays along with DNA/ RNA isolation, protein quantification/ isolation and quantitative PCR. My research focus has shifted over the last few years. Initially, I explored the complex relationship of the TGFB pathway when influenced by the retrotransposon LINE1 and its encoded protein, L1orf1p. I have also been involved in a project that involves the Hepatitis C Virus and its interruption of the DNA damage response even after viral clearance.Due to my work on these projects, I have been published in two reputable journals. - HCV Activates Somatic L1 Retrotransposition—A Potential Hepatocarcinogenesis Pathway- Expression of retrotransposon protein L1orf1p activates TGFβ signalling in hepatocellular carcinoma via cytoplasmic Pin1 upregulationCore Skills: DNA/ RNA extraction, cDNA synthesis, genotyping, cloning, lipoparticle and lentiviral transfection, molecular cloning, target validation, drug screening, qPCR, mammalian cell culture, assay design, western blot, animal handling, dissection/ tissue processing, immunohistochemistry, immunofluorescence, confocal microscopy, protein quantification/ isolation, HPLC, spectrophotometry, GC, FACs, flow cytometry and RNA sequencing & analysis. Assays: Proliferation, migration, invasion, BCA, ELISA, cell viability, IC50Animal handling license - PIL AB Date acquired: 04/11/2021

Molecular Oncology Phd StudentNewcastle University Oct 2022 - PresentNewcastle Upon Tyne, England, United KingdomDesigned and performed experiments to explore early genetic fusion mechanisms in Prostate Cancer.Cultured and maintained multiple mammalian cell lines simultaneously.Performed DNA and RNA extractions, purifications and downstream quantification.Regularly assessed cellular transcript and protein expression via qPCR and Western Blot.Utilised a wide range of microscopy techniques such as immunofluorescence and confocal microscopy to visualise cells and tissue. Advanced techniques such as dual staining, z-stacking and spatial multiomics. Propagated bacterial cultures containing plasmids, purified the resulting plasmid DNA and transfected this into mammalian cell lines using lipoparticle and lentiviral transfections.Performed single-cell cloning to generate a homogenous population from a mixed population of cells.Assessed phenotypic behaviour through proliferation, invasion and migration assays using traditional and novel techniques.Designed overexpression or knockout studies to delineate interaction pathways of various intracellular signalling molecules.Fully analysed numerous RNA Sequencing data sets to assess RNA quality, trim adaptor sequences, align sequencing reads to a reference genome, determine thousands of gene expression values and perform Gene Set Enrichment Analysis. Pioneered new rules and regulations to improve Health & Safety within the laboratory. Supervised and mentored undergraduates and post-graduates where I managed their projects, reviewed their data and proofread their dissertations.Undertook practical demonstrations and led seminars to develop my scientific communication skills. -
