About Daniel Doheny, Phd

I am a recent Ph.D. graduate excited to leverage my strong research background and expertise in assay optimization and preclinical drug development to impact disease on the molecular level and help bring new diagnostics and therapies to the clinic.After studying Biochemistry at UNC Chapel Hill, I interned at The Hamner Institutes for Health Sciences where I learned the basics of cell culture and how to apply the scientific method to develop and validate cell-based in vitro assays. When The Hamner was acquired by SciMetrika and rebranded into ScitoVation, I expanded my working knowledge of assay technologies to include colorimetric and fluorescent plate reader assays, ELISAs, flow cytometry, and high-content imaging. I worked on a multidisciplinary team to develop high-throughput assays to model tissue responses to estrogenic compounds and those thought to cause abnormal adipocyte growth to better predict human responses and inform chemical risk assessment. As my skill set and role within the company grew, I found myself asking technical and conceptual questions that I could not answer with my current knowledge, but knew a career in research was right for me. To continue to advance, I started my graduate education at the Wake Forest Graduate School of Arts & Sciences in August 2017.For my Master’s degree, I joined Dr. Hui-Wen Lo’s lab to identify and study inhibitors of the oncogenic protein tGLI1 which promotes glioblastoma and breast cancer tumor progression and metastasis to the brain by potentiating the cancer stem cell population. Using a cell-based compound screen, we found that ketoconazole, an FDA-approved antifungal, selectively killed tGLI1-expressing breast cancer cells without affecting normal cell populations in the brain and breast microenvironments. This preclinical work laid the foundation for an ongoing early Phase I clinical trial to determine if ketoconazole can modulate tGLI1 signaling in brain biospecimens. Excited to continue learning and grow into an independent scientist, I graduated with my Master’s degree and transitioned into the Ph.D. program under Dr. Lo’s continued mentorship.My dissertation research involved creating and characterizing novel small molecule tGLI1 inhibitors to treat breast cancer brain metastases and recurrent gliomas.
