Structural, functional, and biophysical characterization of TREM2 disease variantsWork from this project has resulted in four publication, including three first-author publicationsGoal: To understand how disease variants in TREM2 effect the protein's structure and function. By understanding how these variants alter TREM2 function, we will also learn more about this important but poorly understood innate immune receptor. This knowledge will enable rationally designed molecular therapies to selectively rescue TREM2 function in disease. Methods:X-ray crystallography, biophysical assays of protein folding and stability, flow cytometry, cellular signaling, protein-ligand assays, recombinant protein expression and purification (mammalian and bacterial), molecular cloning. Results: Determined the crystal structure of the hTREM2 Ig domain.Demonstrated that variants linked to different disease have distinct impacts on TREM2 function.Identified sulfated proteoglycans as a cellular ligand for TREM2 and found disease variants alter binding to this ligand.Used loss- and gain-of-function variants to identify a ligand-binding surface on TREM2.Studied the interactions between TREM2, phospholipids, ApoE, and sulfated glycosaminoglycans.Other work in my thesis lab has resulted in two additional mid-author publication
