The AMD Lab's GENCOV project assesses genetic variants of 1,500 COVID-19 patients to identify geneticvariants that influence COVID symptom severity. In this role, David has:• Filtered and analyzed genomic data, and reported information to research participants with COVID-19. • Classified patients’ genetic variants in accordance with ACMG guidelines.• Utilized genetic databases including Clinvar, gnomAD, UCSC, OMIM, GenCC, and ClinGen to generate comprehensive genetic classification reports.• Outlined association studies between patient HLA and serology data, and viral lineage of SARS-CoV-2 to be conducted over the course of 2021.

In this role, David has:• Cloned genetic material encoding proteins using PCR techniques and created recombinant plasmids using restriction enzymes and DNA ligase.• Transformed plasmids containing protein coding genes into both Pichia pastoris yeast and Escherichia coli bacteria using electroporation techniques.• Created media, buffers, and used microbiology spread plate techniques to grow colonies of single celled organisms.• Used Pichia pastoris yeast and Escherichia coli bacteria to express, purify and isolate MGAM, SI and BT_3302 enzymes for activity assays and crystallization.
