I conducted research towards understanding cellular signaling mediated by protein kinases and protein phosphatases using chemical biology. Protein kinases and phosphatases control the phosphorylation of proteins dependent upon external signals that a cell receives. These pathways control many different aspects of cellular function from their life to their growth and even their death. When these pathways malfunction then many diseases can occur such as cancer, autoimmune and neurological diseases. Thus understanding signaling is important from basic biology to human health. I specifically developed orthogonal, allosterically regulated kinases and phosphatases to use as tools for investigating cellular processes, which cannot be addressed using either biological or chemical methods alone. The project involved the design and engineering of proteins using known protein-peptide interactions to inhibit and subsequently activate our peptide-inserted kinases and phosphatases in vitro and in cellulo.
