He received his PhD in 2000 from the University of Rouen and then conducted a post-doc at the National Institute of Mental Health in Bethesda. He received his PhD in 2000 from the University of Rouen and then conducted a post-doc at the National Institute of Mental Health in Bethesda. In 2002, he was recruited as INSERM Researcher and since 2008, he is the head of the team ‘Neuropeptides, Neuronal death and Cell plasticity’ at Inserm U1239. His project is focused on the functional evaluation of the genes and proteins that are regulated by peptides during brain development or in adult pathologies such as Parkinson and Stroke. David Vaudry is also scientific director of the PRIMACEN cell imaging platform (http://www.primacen.fr) and deputy director of the PISSARO proteomics platform. David Vaudry has published 146 research papers referenced in Pub Med (5402 citations according to ISI Web of KNOWLEDGETM, h-index 36) in the fields of Neuroscience and Cell Signalling in International journals including Journal of Neurochemistry, Molecular Neurobiology, PNAS, Science and Pharmacological Reviews. From 2012 to 2015, he has been General Secretary of the French Neuroendocrine Society and he is currently the coordinator of the LARC-Neuroscience Network (http://larc-neurosciences.org). My objective was to identify neuropeptides which control neuronal cell death and plasticity, and to characterize their mechanism of action in physiological and pathological conditions, in order to evaluate their therapeutic potential for treatment of complex brain insults such as perinatal hypoxia or stroke. Through genomics and proteomics approaches, we identify neuropeptides regulated by hypoxia during the developmental period or stroke at adulthood, and investigate their functional… Show more My objective was to identify neuropeptides which control neuronal cell death and plasticity, and to characterize their mechanism of action in physiological and pathological conditions, in order to evaluate their therapeutic potential for treatment of complex brain insults such as perinatal hypoxia or stroke. Through genomics and proteomics approaches, we identify neuropeptides regulated by hypoxia during the developmental period or stroke at adulthood, and investigate their functional implication. We not only characterize peptides which are efficient to prevent neuronal death in the acute stage but also focus on molecules that promote the delayed neurorepair and regain of functions. Through development of new analogs and galenical approaches, we improve receptor specificity and targeting of our peptides of interest (focusing for the moment on PACAP and ODN) for efficient neurorehabilitation. As peptides are rapidly degraded in the blood circulation, we test some new routes of administration. We in parallel address the innocuity and feature of each kind of administration by monitoring physiological parameters such as blood pressure, heart rate, insulin levels, corticosterone release, cerebral blood flow and cortical activity. In addition to behavioral tests, we investigate by 2-photon microscopy the impact of neuropeptides on neuronal activity and plasticity over the weeks following cerebral artery occlusion. Preliminary data show that daily administration of ODN, started 3 days after the stroke onset, a time point far beyond the established time window for therapeutic interventions progressively promotes functional recovery. Based on recent data obtained with ES cells expressing PACAP, showing the robust plasticity of the microglia even 3 days after the stroke onset, we will also investigate the consequences of this therapeutical approach on functional recovery, inflammatory response and neuronal plasticity.
