Identifying the Biological Function of Cholesterol 25-Hydroxylase (CH25H) CH25H catalyzes the hydroxylation of cholesterol producing 25-hydroxycholesterol, an oxysterol that is a potent regulator of cholesterol biosynthesis and other cellular functions. As a postdoctoral fellow, my project was to elucidate the biological function of CH25H. I found that CH25H was induced in immortalized and primary macrophages when these cells were challenged with pathogens that activate toll-like receptors. A line of CH25H knockout mice exhibited elevated immunoglobulin A (IGA) levels versus wild-type controls suggesting that 25-hydroxycholesterol might be an immunomodulatory sterol. To test this idea, I established an in vitro system in which naïve B cells are stimulated with cytokines to switch production of immunoglobulins from the IgH/IgM class to to IgA. Experiments with this system showed that 25-hydroxycholesterol potently inhibited class-switch recombination (CSR) by naïve B cells by abrogating the transcriptional induction of activation-induced cytidine deaminase (AID), an enzyme required for CSR. The inhibition of 25-hydroxycholesterol was specific for B cells and did not inhibit T cell proliferation or T cell secretion of cytokines. My data indicate that activation of the innate immune system through macrophage toll-like receptors leads to the synthesis and secretion of 25-hydroxycholesterol, which in turn regulates the production of immunoglobulins by B cells of the adaptive immune system.
