Assays used in my group include flow cytometry immunophenotyping, intracellular signaling and cytokine production, MSD chemiluminescence, bulk and single cell RNA-Seq, immunofluorescence, RT-PCR, and various functional cell assays. • Lead scientific efforts for IL-12 partial agonist (STK-026) which avoids NK-mediated toxicities but retains anti-tumor efficacy and activity on activated T cells. Analyze and interpret data, author pre-IND, study reports, conference posters, and scientific manuscript. • Support clinical biomarker flow cytometry design and data analysis for IL-2R alpha-biased IL-2 partial agonist STK-012 and STK-009/SYNCAR-1 Orthologous IL-2/CAR-T programs • Support (JRC member) Merck collaboration that successfully delivered lead and backup humanized Hu/Cyno XR Surrogate Cytokine (VHH) Agonists for autoimmunity which blunt inflammatory Th17 responses and inhibit colitis• Prepare and present pre-clinical data in BD discussions resulting in $40+M deal with Sanofi for engineered IL-10 agonists for autoimmunity designed to avoid T cell activation and resultant anemia but keep immunosuppressive functions.
