    Ph.D. Thesis : "Cloning, Purification and Characterization of Vibrio cholerae's Cell Division Accessory Protein FtsA and Development of Potential FtsA Inhibitor(s)". Under the Guidance of Dr. Gopal Chakrabarti, Associate Professor, University of Calcutta.Research work included cloning and purification of bacterial divisome proteins FtsA, a membrane protein, and FtsZ, a cytosolic protein, along with their recombinant mutants, from Vibrio cholerae. Interaction parameters for these two proteins, and also their several mutations (deletion mutants and SDM mutants), were determined in-vitro using several biophysical techniques. Localization studies of these proteins were performed in-vivo. Polymers of both the proteins were studied and visualized both in-vivo and in-vitro using different imaging techniques. Several small molecules were screened against FtsA in an attempt to halt cell division of Vibrio cholerae. Binding site of Morin, a bioflavonoid, found to inhibit polymerization of FtsA and subsequently the cell division of Vibrio cholerae, was extensively studied with in-silico docking and mutational studies.Also worked in simultaneous projects involving screening of small molecules on animal cancer cell lines (lung cancer and cervical cancer) and in-vitro studies on their anti-cancer activities. In-vivo studies were also performed to monitor their effect on various cellular pathways affecting cell survival. Processes like Autophagy, Apoptosis, Cell cycle analysis, cancer stemness properties were monitored. In-vivo protein expression levels were monitored by western blot and mRNA levels for gene upregulation and downregulation were monitored. Effect on Microtubule cytoskeleton assembly was studied. Imaging of animal cells to localize different fluorescent tag proteins and organelles was performed. Small molecules were docked on proteins to study the binding pockets. Pre-clinical experiments with potent small molecules on mice model were done.
