Weber's, PhD projects.In the summer of 2006 I was awarded a National Science Foundation Research Experience for Undergraduate's summer internship at the American Museum of Natural History under Dr. Rob DeSalle, PhD. Weber, PhD I assayed the reliability of a Denaturing High Performance Liquid Chromatography wave nucleic acid analysis system in detecting haplotypes of MHC Class II alpha and beta loci. I also enjoyed cloning sequences using vectors and competent E. coli. I analyzed data by aligning DNA base pairs using MacClade™ and ClustalX™ software and employed the use of geneDoc Sequencher™ software for DNA sequence modification. Ultimately this research investigated the effects of global warming on arctic animals and marine mammals by examining the major histocompatibility complex (MHC) class II of DNA that is involved with immunity in all vertebrates. This data resulted in a publication on polar bear genetics. -
Summer Internship From The National Science Foundation'S Research Experience For UndergraduatesMote Marine Laboratory May 2005 - Aug 2005Sarasota, Fl, UsIn the summer of 2006, under the supervision of Dr. Kim B. Ritchie, PhD, I worked on genetic fingerprinting of coral aquaculture. This internship consisted of intense troubleshooting for running primers with GC clamps (extra Guanine and Cytosine bases at the ends of DNA which lend to their strength) through PCR with specific microsatellite markers on a Denaturing Gradient Gel Electrophoresis (DGGE). Primer clamps are tricky in making the PCR annealing step troublesome, but are needed for this specific gel electrophoresis in the denaturing process. The silver nitrate stained bands will be used to fingerprint coral nubs. This was not discouraging as I found troubleshooting to be fascinating and overcoming problems is an accomplishment I appreciate. I also, snorkeled for coral mucus samples and coral fragment samples in the Florida Keys to detect present symbiotic bacteria. Dr. Ritchie trained me in coral genetics as well as in marine microbiology. I learned to Inoculate and streak diluted mucus samples on agar plates. I helped maintain strains of marine bacteria. I organized and maintained the microbiology laboratory by disinfecting and cleaning. I generally prepared and dispensed biological media and various buffers appropriately. Lastly, I handled and disposed of hazardous and non-hazardous materials in accordance with lab safety protocols.
