As a member of Manipal BioMachines, I contributed to a synthetic biology project focused on enhancing lipopeptide delivery using a bacterial systems. My role spanned multiple domains, including research, design, analysis, and public engagement, to ensure the project’s success. Key contributions included: • Research & Development: Conducted in-depth research on alternative lipopeptides, their molecular mechanisms, and potential release circuits. Shortlisted bacterial strains with the necessary operons for lipopeptide synthesis, documented production metrics, and optimized promoter sequences for enhanced efficiency in engineered bacteria. • Wet Lab Collaboration: Designed and maintained protocols for promoter testing, developed Gibson assembly fragments and primers, and supported wet lab teams in testing and optimizing sequences. • Computational Analysis: Performed docking studies to validate sequence modifications, ensuring functional integrity of promoters and operons. Compiled and evaluated a comprehensive list of promoter options for lipopeptide production. • Documentation & Data Curation: Authored the “Promoter Handbook,” a detailed resource on constructing and selecting promoters for enhanced functionality. • Visualization & Media: Created animations, graphics, and content for the team’s wiki, social media, and educational outreach. Played a key role in designing the project logo and producing the promotional video and audiobook content. • Public Engagement & Outreach: Led science communication sessions with biotechnology and synthetic biology professionals and organized outreach activities targeting the agrotech sector. • Technical Skills: Assisted in coding for the team’s wiki, ensuring a seamless online presence.
