About Dennis Phan
I am a research professional specializing in upstream process development with additional experience in LNP process development and CHO cell line development. I currently work to define parameters for adherent and suspension cell-based processes in an early-stage biotech environment. Before that, I was involved in making LNP batches at bench scale to establish platform parameters, as well as conducting tech transfer to GMP manufacturing. I also worked with adherent cell culture for oncolytic viral production and have done exploratory work investigating upstream technologies for Phase 2 scale-up. My previous experience includes monoclonal antibody production from transfection through fed batch to bench-top purification. I have a passion for the cutting edge, a good math problem, troubleshooting via outside-the-box solutions, and streamlining day-to-day lab operations.

Senior Research Associate
CurrentUpstream Process Development: support team in conducting studies for process definition and scale-up of material generation for novel gene-editing delivery platform.• Led study of HEK293T-derived adherent cell line growth comparison on HYPERFlasks and CellSTACKs to evaluate implementation in seed trains.• Compiled historical data from 8 months to identify and trend adherent cell line PDT.
Associate Scientist Ii
In LNP Process Development performing studies for robustness and performing tech transfer for ONCR-021 program.• Conducted studies and consolidated historical data on LNP platform to troubleshoot clarification performance, demonstrate robustness by reducing pump pressure variability, and characterize LNPs by measuring particle size, encapsulation, and polydispersity.• Adapted bench-scale protocols to GMP master batch records for 100x scale-up of LNP platform for in-house manufacturing capabilities, including authoring batch record of Sartorius S1000 TFF skid operation. Collaborated with QA, manufacturing, and MS&T to align workflows.• Operated Masterflex L/S pumps, Watson-Marlow 530 pumps, DynaPro (dynamic light scattering). Made LNP batches at bench scale. Maintained Excel files, recorded and tracked data.• Piloted Cytodex 1 microcarrier work to assess as a potential scale-up technology, performing proof-of-concept, recording cell attachment, comparing cell count methods, and achieving over 4x Phase 1 viral titer before transitioning to LNP Process Development.
Associate Scientist I
Worked in Upstream Process Development designing studies for scaling up production of ONCR-177 platform.• Learned and operated iCELLis Nano as an evaluation of iCELLis 500 as a potential scale-up technology for Vero adherent cell culture for Phase 2, observing up to 4x Phase 1 viral titer on the bench. Coordinated with Pall PDS to conduct investigatory work and proof-of-concept.• Maintained seed trains and operated CellSTACKs to produce ~6 x 109 cells on a biweekly basis to supply Downstream Process Development while also performing cell-based assays to characterize viral titer.• Supported GMP process with supplemental experiments for trouble-shooting, served as Person-in-Plant for GMP manufacturing on key upstream operations, and assisted GMP batch record review.• Performed regular Vero cell culture on T-flasks, CellSTACKs, and HYPERstacks.• Operated and maintained Leica DM IL LED, ChemoMetec NucleoCounter NC-200, Vi-CELL XR, NOVA BioProfile FLEX2. Maintained electronic lab notebooks, Excel files. Prepared and presented presentations at internal meetings.
Research Associate I
Worked in Cell Line Development and Upstream Processing to maintain monoclonal antibody cell lines. Analyzed and presented project data to clients, conduct fed batch studies, and maintain single-use bioreactors.• Designed media condition study to troubleshoot lack of growth of single cell line, achieving active cell division of over 80 cell lines and resuming project progress after 3-week loss of productivity.• Prepared experiment to evaluate and compare evaporation rates of different incubators, identifying evaporative losses of 2 mL per week that potentially affected cell cultures.• Wrote standard operating procedure (SOP) for running SDS-PAGE, establishing uniformity in SDS-PAGE production across multiple departments (Upstream, Downstream, and Cell Line Development).• Implemented registration system to record gas and liquid nitrogen supply maintenance, assessing consumption patterns and reducing waste caused by premature resupply.• Produced cell lines by performing limiting dilution to achieve monoclonality, selecting monoclonal lines via Solentim, and scaling them up to shake-flask scale.• Maintained shake flasks for fed batches, compiled cell growth and protein production data in Excel and PowerPoint, worked with client to optimize protein capture during benchtop purification. • Operated HPLC to perform Protein A chromatography to record mAB titer.• Obtained yearly GMP training for operation of GMP-critical equipment and facilities.
Life Sciences Intern
Research Assistant
Studied enteric pathogen Shigella flexneri in the Mucosal Immunology and Biology Research Center under Christina Faherty, PhD. Performed protein analysis on proteins OspE1 and S0162 via co-immunoprecipitation and western blot.• Designed study to visualize colocalization of OspE1 and S0162 in Shigella. Purified cell membrane fractions to isolate protein. • Determined optimal capture methods for OspE1 and S0162 as anti-His and anti-flag agarose. Redesigned cell growth conditions and centrifugation methods based on western blot results.
Senior Instructor
Taught mathematics to students in grades 1 through 8 with Mathnasium’s intuitive model. Learned to adapt teaching style per student to illustrate concepts in a tailored manner.• Oversaw instruction of over 30 students, tracking their progress through curricula and ensuring 100% concept retention through biweekly evaluations. Increased membership acquisition through one-on-one trial lessons.
Summer School Teacher
As a summer school program teacher, taught classes of rising 1st, 2nd, and 3rd graders. Developed full lesson plans to reduce "summer slide" based on Boston Public Schools' mathematics requirements for grades 1 through 3. Supervised groups on weekly field trips.
