The main project of this PhD is focused on using a neutrophil-stimulating, bi-specific antibody as a vehicle for organic, immuno-active payloads in order to treat a variety of cancers in pre-clinical models. This involves molecular techniques such as bi-specific antibody design/production, genetic cloning, binding- and ELISA assays; as well as functional cellular techniques such as Antibody-Dependant Cellular Cytotoxicity/Phagocytosis (ADCC/ADCP) and chemotaxis assays. Passing these in vitro tests will lead to possible candidates being tested in a pre-clinical tumor mouse model. In the group of Prof. Dr. Marjolein van Egmond at the department of Molecular Cell Biology and Immunology (MCBI) situated at the Amsterdam UMC, location VUmc.

Department Of Human Genetics

During this internship I worked on fundamental research into the role of HMGN1 and HMGN2 in transcription-coupled nucleotide excision repair (TC-NER). This included, but was not limited to, generating knockout cell lines via CRISPR-Cas9 mediated gene-editing, performing functional assays specific to TC-NER, investigating protein-protein interactions via immuno-precipitation and live-cell imaging.Under supervision of PhD candidate Katja Apelt in the group of Dr. Martijn Luijsterburg.

Responsibilities included preparing the experiments and instructing students on how to perform them. Giving a small lecture at the end to explain everything they have done and should have learned.

Massachusetts General Hospital, Center For Transplantation Sciences

During this internship I primarily worked on translational research into the role of recipient- and donor-derived exosomes in solid organ transplant tolerance involving a murine mixed bone-marrow chimerism model. As a secondary project, I performed a retrospective study to identify risk factors for post-transplantation lymphoproliferative disorder (PTLD) in non-human primate (NHP) mixed chimerism transplantation models.Under supervision of MD. Kortney A. Robinson in the group of associate professor Gilles Benichou.

Department Of Vascular Surgery

During this internship I worked on co-stimulation of T-cells in a murine vein graft disease (VGD) model. Specifically, I looked at the effect of CD70, CD80/86, and CD70/80/86 knockout on the development of VGD, measured in patency of the vein graft as well as macrophage infiltration and collagen content in the vessel wall.Under supervision of PhD candidate Karin H. Simons in the group of Prof. Paul H.A. Quax.
