My contributions to drug discovery at Pfizer drew from my expertise in ADME optimization, synthetic methods, medicinal chemistry design strategies and computational chemistry. The joint appointment to both medicinal and computational groups gave me a unique perspective and approach to lead finding and optimization. My deep knowledge of ADME and in-silico filtering provided Pfizer with novel properties screens and improved the quality of their screening collections.In the Antibacterial therapeutic department, I conducted HTS Triage analysis for antibacterial screens and successfully lead multiple AB programs seeking novel target agents, including natural products.At Pfizer Groton, I provided full-time computational chemistry support for programs in the CV therapeutic area.

Parke-Davis recruited me to explore novel kinase targets in the Oncology therapeutic area. There we discovered the first example of a deep pocket-inhibitor. I applied my knowledge of kinase inhibitor design and SBDD strengths to develop novel, potent inhibitors.

My work at Novartis-Summit focused on cardiovascular disease, where I developed an orally biovailable long-acting protease inhibitor for Endothelin Converting enzyme. I led the chemistry strategy for a kinase program addressing post-MI reperfusion injury. My interest in compound physical properties and ADME led me to introduce a simple approach to solubility determinations, later dubbed "kinetic" solubility. My contributions in meeting project milestones and introducing new approaches resulted in my advancement in Novartis.

Sterling Winthrop hired me to develop a biological using the then-novel technology of pegylation. I developed the analytics and scale-up technology to pegylate protein for enhanced in vivo exposure and this compound moved into clinical studies. I developed here my expertise in protease inhibition strategies with the development and disclosure of Caspase-1 inhibitors.
