We (at the research lab in work) have two fluorophores specific locus on the Dirofilaria immitus genome. Once both the fluorophores hybridize (specific only for D.immitus) it gives a specific color under the microscope. We have been studying various parameters like concentration of DNA or primers, hybridization conditions, sensitivity and specificity of the system. -

So CRP chimeras where created with known ligand binding domain and unknown DNA binding domain and vice-versa. They were sent for physiological analysis of genes activated by transcriptional activation.Inter-species CRP complimentation could be a difficult task because high CRP activity is lethal. The hypothesis is that the high transcriptional activation of certain genes causes death of bacteria. This lead us to randomize two specific amino acid positions on the CRP protein creating a range of CRP activity.
