I worked on a project studying the control of immunological synapses in regulatory T cells (Tregs). The role of cytoskeleton regulators in immune synapse formation of Tregs was studied through generation of knock-out of candidate proteins (cytoskeleton proteins involved in molecular transport) in Jurkat T cells and murine CD4 T cells via CRISPR/ Cas9 technologies. Techniques learned: Cloning, Lentiviral Transductions, gene editing via CRISPR/Cas9, Immunofluoresence, mice handling and prepping for ex-vivo experiments, immunophenotyping via FACS, in-vitro Th differentiation cultures (Th1, Th2, Th17 and iTreg)

Project: To assess the effect of Ayurvedic Treatment on systemic immune response and oxidative stress in oral cancer patients treated with radiotherapyThe project was funded by ICTRC, Wagholi Pune to study the immune profile of oral cancer patients who underwent radiotherapy. Peripheral blood samples were collected from patients and control group at various treatment time points were analyzed for T cell compartment by FACS to investigate the immune profile of the patients. The CD4 and CD8 T cell populations were sorted and their proliferation status was evaluated by functional assays.

I worked on a project understanding the epigenetic regulation of immune responses of human gamma delta T cells. The role of Ezh2 methyltransferase and Histone deacetylases in modulation of effector functions of gamma delta T cell was studied. I accquired hands-on training in isolation of primary T cells from peripheral blood lymphocytes and in-vitro expansion of Human gamma delta T cells as well as immunological techniques such as FACS, Immunoprecipitation and ELISA.
