    The amyloid-beta peptide accumulates in the brain of Alzheimer’s disease patients and initiates disease onset. Our lab studies the cellular pathways that regulate the formation and clearance of amyloid-beta. I am the lead technician performing in vivo microdialysis in mouse models of AD; this technique enables us to study how amyloid-beta changes every hour over several days in response to genetic, drug, or behavioral manipulation. The amyloid-beta microdialysis technique was originally developed in our lab and is only used by 7 other groups in the world; including Case Western Reserve and various other universities who visited Washington University so I could teach them this technique. In addition to performing our own experiments, I am also responsible for organizing, coordinating, and executing research projects in collaboration with academic and corporate groups. Techniques used: In vivo Aβ microdialysis, small animal stereotaxic surgery, intracerebral injections, drug administration to mice, sandwich ELISA, custom construction of Electroencephalography probes, and data analysis.

    Huntington’s disease derives from a polyglutamine expansion in the huntingtin gene. This expansion results in the misfolding and aggregation of the protein in the cell where it becomes neurotoxic. Accurate quantification of the protein in in vitro is difficult. I constructed a huntingtin protein with a Cyc-Cys-Gly-Gly motif attached to either the amino or the carboxyl terminal of the gene so that aggregates could then be labeled using the biarsenical reagent FlAsH-EDT2. This reagent binds only to a tetracysteine motif which the huntingtin aggregates reconstituted, coordinating with FlAsH-EDT2 and fluorescing. Wild type huntingtin, which does not aggregate, did not fluoresce. The microbes are identified by sequencing the 16s ribosomal RNA, chosen for its stability and conservation between different species of bacteria and Achaea. I prepared samples for sequencing through amplification and purification of samples, which I then used in transforming E. coli competent cells.
