At Rubius I lead the protein sciences group.At Novartis I was on projects in various disease areas that have involved compound target ID to early drug discovery efforts. In this position I worked on assay development (TR-FRET, SEC, FP, DSF, LC-MS, SPR, SEC-TID), protein design (designing protein sequence), protein purification (purified thousands recombinant proteins with various tags), protein biophysics/analytics, protein array technology, and protein/small molecule screening. I worked at Novartis (NIBR) in the Chemical Biology and Therapeutics department in the Chemical Biology hub.Bristol Myers Squibb (Adnexus) I was part of the protein sciences group. In that role I purified thousands of His-Tagged and untagged recombinant proteins across multiple programs to support high-throughput screening, assay development, biophysical and biochemical characterization.o Assay development: Time resolved fluorescence energy transfer (TR-FRET), LC-MS for size exclusion chromatography-target identification (SEC-TID), differential scanning fluorimetry (DSF), surface plasmon resonance (SPR), fluorescence polarization (FP), colorimetric and fluorescent assay formats.o Protein Expression systems: Escherichia Coli (E. Coli), Mammalian Cells (HEK293, CHO).o Protein purification, characterization, and quantification techniques: AKTA, SDS-PAGE, Analytical SEC, Nano Drop, UV- Spec, LC-MS, DSF, DSC, SEC-MALS.o Chromatography (pre-packed and self-packed): affinity, hydrophobic, ion exchange, size exclusion, custom resins.o Purification workstations: AKTA various models (GE), BioCad (Applied Biosystems), Profina (Bio-Rad), gravity (manual)o Protein refolding experimentso Quantification: BCA (bicinchoninic acid) assay and extinction coefficient based UV-VIS spectroscopy, LAL assay.o Characterization: SDS-PAGE, Western Blotting, size exclusion chromatography, LC-MS, DSC and DSF (thermal melt assays).o Automation: Beckman BioMek, GXII, LC-90, Sciclone LH3000, Aria, Masslynx.
