I also led two other projects (a) Phosphoproteomics interaction project, wherein possible effect of SNP in BRI1 kinase domain on downstream signalling was dissected upon. (b) Cloning and Functional validation of leucine rich repeat receptor kinase for its role in Fusarium resistance through VIGS (Virus induced gene silencing), expression and signalling analysis. I supervised a team of 23 professionals including Marie-Curie ITN PhD, Masters and undergrad students. I led review meeting that look into the Strategic and scientific hypothesis, its rationale, providing full support on areas such as planning, design and realistic scientific accomplishments, future experiments accuracy, supervision & training, Lab aseptic and equipment review, Lab ordering/POs, Collaborators networking, Lab safety & Occupational Health, project review report, SOEs/SOPs, Ethics and dissemination and Resourcing.I could successfully complete all of the 3 projects, developed resource of 2016 GENOME EDITED validated lines of BRI1 receptor kinase snd revealed differential regulation of BRI1 signaling due to SNP in BRI1 kinase domain. I validated the function of LRR-receptor kinase by reverse genetics approach and got European patent application no.: EP17160440.8 for first wheat Fusarium resistance LRR receptor kinase. I successfully disseminated and published papers in peer high impact factor journals. I managed team of diverse culture and background. I established strict GMO guidance of aseptic rules, ISO17025 and applicable GMP. I introduced a new inventory and lab Management System for quick access to chemicals and stocks.

Role Overview:• Working as main lead scientist of the research project entitled “Expression of N15 labelled HIV1 protease for development of drugs” and to train the technicians, students, interact with collaborators and manage lab logistics to achieve the scientific goal• I need to bioengineered molecular expression vector for the N15 labelled wildtype along with mutant variants of HIV-1 protease from Multi Drug resistance (MDR) patients. Once expressed, the HIV-1 protease was needed to be purified and structurally characterized by NMR, IR spectroscopy, MALDI-TOFF and LC-MS to reach at possible functional inhibitors to facilitate drugs development, even for MDR patients. • My role also incorporated a framework of Genetic manipulated organisms (GMO) protocols and training, experimental Planning, Graduate & post grad teaching, lab management, lab review meeting, supervision and dissemination. • Supervise and guide a team of project fellows and masters/undergrad students for their relevant thesis research workKey Achievements:• Secured 0.5 M RANDS Grant funding for this UKZN-PDF project in 2013.• I successfully tackle the nagging problem of expressed proteins going into inclusion bodies by adjusting the culture parameters and even successfully harvesting inclusion bodies to rescue complete expressed HIV-1 protease.• I formulated and successfully designed the scientific strategy while keeping in mind of learning focus of students to expressed and purify N15 labelled recombinant HIV-1 protease. The N15 labelled HIV-1 protease was purified and characterized by MALDI-TOFF and LC-MS for inhibitor development• I developed my skills of proteomics, protein structural characterization, enzyme assay and bioengineering techniques.• Undergone GMO training under strict guidance of aseptic rules, ISO17025 and GMP guidelines.

(d) Molecular biological analyses of Brassica plants over- expressing the tobacco basic 1,3 chitinase gene and bioassays of the transgenics using Alternaria brassiceae to check their fungal resistance. • I had full responsibility for activities relating to research work in lab, Greenhouse, Bioassay experiments, lab orders Management, Training, Radioactive experiments.Key Achievements:• Successfully created Suppression Subtraction Hybridization (SSH) salt stress cDNA library, identified several promising salt stress associated genes like UBC4, eIF2α, PPiase, E3 Ubiquitin ligases and validated in corresponding yeast mutants. These genes were functionally validated by overexpression in rice and tobacco plants. • Characterized pyramid HSP100 and HSP18 transgenic lines. I isolated root specific promoter from rice and analyzed it by promoter-GUS assay and EMSA. Brassica overexpressing lines were also phenotyped and characterized for fungal resistance. I developed and learned advance skills of molecular biology and learn time and research management of multiple projects.
