    I conducted research to investigate the effects of anti-diabetic incretin-based therapies upon the microvasculature; utilising a cerebral endothelial cell (EC) line. I acquired a broad range of critical skills for conducting effective self-directed research and competence in an array of biological investigative approaches. I carried out considerable cell culture with endothelial and monocyte cell lines and isolated primary leukocytes.My PhD focused on three areas: EC proliferation, nitric oxide production and inflammation. Proliferation was examined through BrdU, Trypan blue, Cyquant and WST-8 experiments. I went on to investigate downstream mechanisms of action behind observed proliferation. I characterised the ECs, confirming the presence of the GLP-1 receptor through immunocytochemistry and western blotting. Inflammation and cell viability experiments were conducted utilising, flow cytometry and cell surface adhesion molecule ELISAs. NO production was assessed via eNOS phosphorylation, an NO probe and the measurement of nitrite and nitrate products using the Sievers NO analyser. As part of the Medical School I was also member of the 'Endothelial Cell Biology Group', in which we collaborated in weekly meetings and discussed ongoing project findings. I was in charge of facility's maintenance and ordering of laboratory gases and ethanol for cell culture. For this role I completed the 'BOC safe handling course'.

    I very much enjoyed the hands-on care of patients whilst working in healthcare. This bolstered my interests in medical and clinically translatable research. The nature of the role as an HCA is caring for the hygiene, nutritional and supportive needs for all patients under my care for the shift. whilst ensuring that you remain vigilant to the condition of everyone under your care; quickly reporting any marked changes to your senior on the shift. A single HCA and two nurses may be overseeing 12-16 patients at one time so it was imperative to work autonomously. It was essential to have good time management, confidence and to be proactive as patients conditions and states of mind, at times change rapidly. At the Royal United Hospital I was stationed upon a secure unit for the "Care of the Elderly" this meant that I met a large number patients that were at the extreme end of their disease and therefore the symptoms were very complex at times to deal with. This position was very positive for me. I gained a great deal of experience dealing with heavy work loads, coping with night shifts and difficult situations which often required delicate communication with confused people.
