Adequately designed primers were used to distinguish between methylated and unmethylated genes by Methyl specific PCR. mRNA expression analysis of ATM,TP53,GSTP1,APC and TIMP3 by RT PCR was also done. The findings indicated that there was hypermethylation of promoters of ATM,APC,TIMP3 and GSTP1 and this could serve as biomarker in distinguishing the cancerous from non-cancerous cells. Also, hypermethylation of three of these, namely, ATM, APC and GSTP1, was associated with aggressive form of cancer lymph node positive phenotype and metastasis. The ATM and APC promoters were suggested to be useful as biomarkers. Correlation with age was also observed and the data was statistically found to be significant.This work leads to my PhD degree with 3 research publications in peer reviewed journals and 3 conference presentations. -
Assistant ResearchPrinceton University Jun 2010 - Sep 2010New Jersey, UsaFrom large collection of cultivated denitrifiers, I recovered culture from liquid N2 storage and cultivate them using standard microbiological technique. I also helped to isolate additional denitrifier strains from sediment samples, in order to add them collection. I isolated DNA from the cultures and screened them for the presence of nir S gene (encoding nitrate reductase ) using the polymerase chain reaction (PCR).I also helped to extract DNA from sediment samples, which is more challenging than extracting DNA from cultures. I performed several different PCR assay on these samples. In addition, I helped run some nutrient assays, measuring inorganic nitrogen concentration in seawater samples using spectrophotometric methods. -
