Development of computational structural models for GPCR based on homology modeling for identification and characterization of specific receptor interaction pattern resulting in receptor modulation. Specific GPCR allosteric modulator design by structure based and pharmacophore based design.Design and development of Ion channel inhibitors by structure based and analogue based design.Design and identification of small molecule candidates with dual/selective enzyme inhibition profile.Identification and design of new small molecule enzyme activators based on structure based and virtual screening of database. Characterization of the protein-protein interaction pattern by molecular dynamics simulation to identify and design small molecule inhibitors. Working in close association with the team consisting of medicinal chemist and biologist for the validation of hypothesis to design and deliver new chemical entities for different targets.Handling of molecular high throughput screening (HTS) data to identify hits. Utilization of computation tools for identifying new hits, developing hit to lead and to optimizing lead for wide range of therapeutic targets from various therapeutic areas. Evaluation of drug targets of pharmaceutical interest with drug design point of view.Review and evaluation of different drug design software.Database preparation and maintenance of in-house and vendor database molecules for high throughput virtual screening using chemoinformatic tools.

Thesis: Design of various new enzyme inhibitors as anti-mycobacterial agents using in-silico high throughput screening. For various enzyme targets from mycobacterium (Mtb) inhibitors were designed by using different computational modeling approaches.Construction of structural model of Mtb Glutamate Racemase (GluR) based on homology modeling and comparison of the model with pre-reported model by molecular dynamics simulation and examination of structural stability over old model.Identification of new inhibitors using molecular docking and virtual screening by structure based design. Identification new inhibitors with potential to inhibit Mtb Protein Kinase B (PknB) enzyme by analogue based drug design; pharmacophore model building, virtual screening and docking simulations.Construction of structure based docking model and validation of hypothesis for Mtb Isocitrate lyase (ICL) enzyme inhibitors.In-vitro assessment of ICL enzyme inhibitory activity of the hits identified from virtual screening of database.
