    This involved Molecular characterization of a novel heterozygous splice site mutation (c.2046 + 1) G>T in LRSAM1 gene in a patient with CMT 2 of Autosomal dominant (AD) inheritance during my first lab rotation. A heterozygous pathogenic variant was identified in the gene encoding Leucine rich repeat and sterile alpha motif 1 (LRSAM1) by direct c DNA sequencing of genes within the region in affected c DNA (reverse transcri bed from patient’s RNA) samples. The single nucleotide change mutates an intronic consensus donor splicing site from GG to GT. I isolated the RNA from patient’s blood and converted it to c DNA. The Sanger sequencing indicated the lengthening of the m RNA product by 63 base pairs confirming the prediction by GENE NET software to be true. So I found that this aberrant splicing due to a point mutation causes an obligatory frameshift and further lengthening of protein by insertion of intron of 63 base pairs between exon 25 and exon 26 in the m RNA.
