I developed a method implementing a combination of long-read and short-read next-gen sequencing to measure the activity of nearly 10^5 sensors across 24 different growth conditions simultaneously. This effort marked an early demonstration of precision engineering in biology, showcasing quantified accuracies of engineering efforts. Additionally, using this approach, we identified several novel phenotypes, some of which had not been described prior to this work, that provided insight into sequence-structure-function relationships underlying these phenotypes.In partnership with a multidisciplinary team, we ensured the data from this work met the quality requirements for machine learning efforts. This approach laid the foundation for the emergence of novel machine learning techniques in protein engineering for genetic sensors, as well as more generalizable understanding of protein landscapes.
